Abstract The objective of this work was to evaluate root phenotyping methods and the ideal phenological stage to quantify the root system of fixed and segregating common bean populations, in order to select superior genotypes. The experiment was carried out in two municipalities in the state of Santa Catarina, Brazil, and the treatments consisted of six genotypes, the Shovelomics and WinRHIZO root phenotyping methods, and the V4-4, R6, and R8 phenological stages. The simple lattice experimental design was used to evaluate the following variables: basal root angle, vertical root length, left and right horizontal root length, total root length, projected area, and root average volume and diameter. For all variables, there was a significant interaction between phenotyping methods and phenological stages, showing their influence on root system evaluation. The Shovelomics and WinRHIZO phenotyping methods are efficient in quantifying the root system of common bean plants and show specificity for phenological stages, regardless of the genotype. The quantification of the root system of fixed and segregating genotypes is analogous in both methods. The Shovelomics method is more efficient in evaluating the root system of common bean at the R8 stage, and the WinRHIZO method, at the R6 stage.

Resumo O objetivo deste trabalho foi avaliar métodos de fenotipagem de raízes e o estádio fenológico ideal para quantificação do sistema radicular de populações fixas e segregantes de feijoeiro, para selecionar genótipos superiores. O experimento foi realizado em dois municípios do estado de Santa Catarina, Brasil, e os tratamentos consistiram em seis genótipos, nos métodos de fenotipagem Shovelomics e WinRHIZO, e nos estádios fenológicos V4-4, R6 e R8. O delineamento experimental látice simples foi utilizado para avaliar as seguintes variáveis: ângulo de raiz basal, comprimento vertical das raízes, comprimento horizontal esquerdo e direito das raízes, comprimento total das raízes, área projetada, e volume e diâmetro médio de raízes. Para todas as variáveis, houve interação significativa entre os métodos de fenotipagem e os estádios fenológicos, o que mostra a influência desses na avaliação do sistema radicular. Os métodos de fenotipagem Shovelomics e WinRHIZO são eficientes para a quantificação do sistema radicular do feijoeiro e apresentam especificidade para o estádio fenológico, independentemente do genótipo. A quantificação do sistema radicular de genótipos fixos e segregantes é análoga em ambos os métodos. O método Shovelomics é mais eficiente para avaliar o sistema radicular do feijoeiro na fase R8, e o método WinRHIZO, na fase R6.

Abstract: Yerba mate (Ilex paraguariensis St. Hill.) is an important woody tree in South America, however the production of quality seedlings is a major problem. Microorganisms that promote plant growth have shown to be efficient biotechnologies in the production of seedlings. The arbuscular mycorrhizal fungi (AMF), it establishes symbiosis with several plant species and increases the accumulation of biomass and absorption of poorly mobile nutrients such as phosphorus. Here, we test the effects of two AMF species on growth, root architecture, phosphorus accumulation and gas exchange of yerba mate seedlings cultivated under different phosphorus levels. We used seedlings in a non-sterile soil, inoculated with AMFs Rhizophagus clarus SCT720A and Acaulospora colombiana SCT115A, and non-inoculated (control) treatment under five levels of phosphorus (0; 25; 50; 100 and 200% recommendation). After 90 days of AMF inoculation, plant dry biomass, root architecture and mycorrhizal colonization were determined and after 180 days, the same parameters plus photosynthetic rate, transpiration rate, stomatal conductance and P content were evaluated. AMF inoculation increased shoot and root dry biomass, total root length, root volume. Plants inoculated with AMF showed higher photosynthesis rate. Phosphorus content and mycorrhizal colonization were increased almost three times when inoculated with AMF. Our findings highlight the importance of AMF inoculation for Ilex paraguariensis seedlings production, reducing the time needed in nurseries to enhance tree performance.

ABSTRACT Genetic variability is essential for gains in breeding programs. The cyclic process of progeny recombination is a strategy to raise the chances of selecting better genotype combinations. The objective of this study was to identify superior progenies between common bean genes groups in first recurrent selection cycle for root system traits. Parents of the Andean and Middle American gene groups were hybridized in a complete diallel scheme. Thereafter, the parents and F1 and F2 populations were planted in the field. To establish the base population of recurrent selection (C0), seven segregating populations (F2) with superior performance for root system were selected and intercrossed, resulting in recombinant progenies (C1). To estimate the selection gain, the parents and C0 and C1 genotypes were compared with regard to the following variables: total root length (TRL, cm), projected root area (PRA, cm2), root volume (VOL, cm3) and number of root tips (RT). The difference between genotype combinations indicates the presence of genetic variability and effectiveness of recurrent selection. The mean genetic progress for root system-related traits was 12.9% (TRL), 12.6% (PRA), 12% (VOL) and 11.5% (RT) in the first recombination cycle. The mean phenotypic performance of seven of the C1 progenies exceeded that of their parents for all root system traits. These progenies are promising as base populations of the next selection cycle.

RESUMO A mudança no uso da terra, aliada às práticas de manejo, contribui para as alterações entre as entradas e saídas de carbono do solo. O objetivo deste trabalho foi avaliar a dinâmica do carbono no solo em ecossistemas nativos e plantações florestais de Santa Catarina. O trabalho foi conduzido entre os meses de fevereiro e maio de 2007. As áreas avaliadas foram: Campo Nativo (CN); Floresta Ombrófila Mista (FOM); Reflorestamento de Eucalipto (RE) e Pinus (RP). Foram analisados os estoques de carbono, carbono na biomassa microbiana (CBM); respiração basal do solo (RBS) e quocientes microbiano e metabólico (qMic e qCO2). As maiores concentrações de carbono foram encontradas na FOM e no CN, e os atributos bioquímicos indicam que o CN e a FOM possuem a melhor capacidade de armazenar carbono no solo, entre todos os sistemas de uso da terra analisados. Nos reflorestamentos de Eucalipto e Pinus, os altos índices de RBS, qCO2 e as baixas concentrações de CBM sugerem que esses sistemas de uso da terra perdem mais CO2 para atmosfera do que os ambientes nativos.

ABSTRACT Changes in land use combined with management practices contribute to changes in soil carbon inputs and outputs. This study aimed to evaluate the soil carbon dynamics in native ecosystems and reforestation in Santa Catarina. The study was conducted between February and May, 2007. The following areas were evaluated: Native Field (NF); Araucaria Forest (AF); Eucalyptus (ER) and Pinus (PR) reforestation. Carbon stocks, carbon microbial biomass (CMB), soil basal respiration (SBR), microbial (qMic) and metabolic (qCO2) quotients were analyzed. The highest concentrations of carbon were found in AF and NF, and biochemical characteristics indicated that NF and AR have the best capacity to store carbon in soil among the land use systems evaluated. Eucalyptus and Pinus reforestation showed high rates of SBR, qCO2 and low concentrations of CMB that suggest these land use systems lose more CO2 to atmosphere than native ecosystems.
